YouTube is one of the simplest things to use and yet, many people don't realize how to optimize videos appropriately.
First off, when picking a title for your video, be sure to research your keyword in BOTH Google and YouTube. There are many times when your YouTube video can end up ranking high on Google because the competition on Google is already pretty low and easy to rank for.
You can even use it to rank a video as it's own "micro niche site"
There are many guides here on how to use Google Keyword Tool, so I won't get into that.

Now when you are using the tool, be sure to use EXACT searches in the MATCH TYPE just as you would do in the Google Keyword Tool.
Now, when you have the proper keyword researched, you just need to use that keyword in every aspect of your video. When you have the video uploaded, be sure to include the keyword within these 3 aspects:
Title
Description
Tags
Many people say to use the tags of the top videos there... yes, it might help. But, if you can be more targeted and just use the tag for the keyword you WANT to rank for, it's probably better.
I have personally done a test and if you just have one tag (which is the keyword) as the tag and title and then have a nicely done description with the keyword, it tends to rank high because it's targeted.
Many times, people upload a video with a link, but don't see any conversions. Youtube has it's own video editor which allows you to put in a little comment that overlays your video. USE THAT TO YOUR ADVANTAGE!
You can easily just persuade a person by putting some phrase which makes them want to click the link instead of watching the whole video.
For example, I did so in my video for Halo 4: http://www.youtube.com/watch?v=LhCJgLNwKPk
I get a conversion rate of about 12%, although, I don't get very many clicks due to the video quality (i was testing it out ).
Those three aspects: title, tag, description, are not the only things that will help you get ranked high. It is good if you can get some start up views from anywhere and try to get some likes and comments. I personally outsource this on fiverr.
Be sure to get QUALITY comments, the more engaging comments you have, the better user input you will get and comments will just come your way without you doing anything.
Be sure not to have more likes and comments than the amount of views you have. It could possibly lead to you being flagged and it just makes your video look suspicious. Gradually increase the likes and comments in accordance to your views.

People have been asking a lot about youtube SEO and what they should do. It's simple, do SEO as if you were doing it for your websites. Except, with YouTube, you can use spammy links like forum profiles and it still works out well.
For all those videos in my channel, I outsourced to a fiverr gig that gave 10,000 backlinks (xrumer blast) and usually after a few days I saw a jump in the rankings.
I haven't tested out doing more SEO towards it like link pyramids, wheels, etc. But, i'm sure they work very well.
Think about it though, if you rank high on youtube and build backlinks to your video, it gives you a chance to also rank high for that keyword on Google.
So, once you're ranked up high, you can just sit back, relax, watch the traffic just pull in. Be aware though, YouTube is jumpy. There are days when my videos don't show up on page 1, then the next day it is #1.
Here's a little bonus tip that I found with my links... use Google's link shortener for links in your description. When I had it shortened with Google's shortener, my videos ranked high. But, when I switched them to bit.ly for about a week, they went WAY down in rankings and the day I switched them back, it went back up. I think Google links to see it's own services being used and not the competition
