Indexing a backlink it's EXTREMLY important because if they aren't indexed by google or any other search engine,the backlink it's useless in terms of SEO.
I would say blog commenting it's the best move,but you must add some diversity to your backlinks,else they won't look natural.With diverisity i mean to say,profile links,blog comments,forum posts,signatures,articles etc.

No,on high PR blogs,manual comenting is a must,if you want your links to stay there.
