my main target is to Focus on USA Traffic.so my question is what is the more effective Domain Extension.
Keyword.Com Or Keyword.US or there is no Connection with the Extension.

Personally I would go for the .com

If I am doing a UK facing site, I tend to go for the .co.uk if the .com is unavailable OR if I have the .com for the American facing version.

Can someone outside uk sign up for .co.uk domain?
 
Both are fine. But personally, I'll go with .com when targeting US traffic.
 
If you are going to target the US traffic, then go with .US TLD domain. Because Google gives you more traffic for .US than .COM.
