Google searches keywords, no matter where they are. So if they are not in main content, you can put them in comment and it will help you.

I think you want to mention about blog commenting, forum posting, guest posting etc. Yes, it definitely helps you. I suggest you to create backlink on high PR websites.

Yes keywords in comments will help soo much but not for YouTube videos.

If you are leaving comments or writing content, keywords should always be used strategically. No stuffing or spamming but used contextually, this is always a good thing.

Yes... but use in limit.. like 1-2 times in comment is enough...
