Do tier 2 links have to be niche related?

And is it ok to have unique but a little unreadable content for tier 2?

Content can be unrelated and semi-unique/readable for tier 2s... you'll be fine.

Yes, 100% Safe. But It depend on 1st Tier too. For example if you want 2nd Tier for Bookmarks, or High Quality Web 2.0. I will suggest to build Web 2.0 links or Bookmarks ..Inshort cross Link Type to Each other.
For Web 2.0 1st Tier, build 2nd Tier bookmarks, and If You have Good High Quality bookmarks or Profiles, Build Web 2.0 links. it Total Depend on How You Plan your Link Campaign.

I am Saying That 2nd Tier You can Build Easily, But It all Depend what Type of Links are your 1st Tier and How Good you want your 2nd Tier.

Tie2 links are more about quantity than quality, you can use spun content for tier 2 links.

Tier 1 should be relevant, readable unique content, tier 2 can be relevant, unique (readability doesnt matter), tier 3 can be pure spam as you only want your tier2s indexed well.

For tier 2 it should be at least 50/60% unique and readable, as for niche it should be a mixture not all irrelevant.
