Dude yesterday I built 20,000 PR3+ profile links to a website and second tier sites and wont see a whole lot of difference in the serps. If I get a 10% increase in traffic week in week out I feel I'm doing well. Profile links are great because they are so easy to automate but it's quantity rather than quality with them I'm affraid. 40 of them aren't going to get you rushing to the top. If you are doing it manually why not try to put your efforts into targeting high quality links instead? Email, phone or write to other sites in your niche and ask if they want to exchange links, get your link out there on high authority sites any way you can. One good ink on a PR6-7 page (note. page not PR6-7 site) is worth so much more than 20,000 profile backlinks.
To give you some idea one nofollow link on a exceptional page increased the traffic to one of my sites one hundred fold in a week. Alot of it was due to traffic from the link but the traffic from search engines rocketed as well. That's where you should be focusing your efforts IMO not filling in profiles manually.
