Hey Guys,

When I submit a video response, does it need to be approved by the owner of the video I am responding to? Also, does any significant number of people click through on a video response link?

The user can set whether they review it first or not, so depends on the video. Also click through depends on the type of video and its popularity. I find people are much more likely to click suggested results after a video rather than video responses.
 
By default it is 'by review'. They can manually set it to approve all though.
 
Not all videos are set to auto-approve video responses. Just keep looking for videos which are set to approve automatically when a video response is posted. It involve a bit of work as most videos are set to manual approval by default.

Depending on the video you're responding, your video can get a good amount of clicks. If you have good and relevant video, chances are the owner of the video you're responding will approve it. Also, keep in mind that many videos in youtube that have lots of views but the uploaders are not active and not online for a year or so, already.
 
Yes, sometimes you see video's taken off if they have not got authrisation
