Will changing ONE frame make the video unique?

If they're using a global checksum it will. If they're hashing every 10 seconds and compare those you will get 9 identical and one different checksum for a 100 sec video.
You can always convert to another format, cut a second here and there and reconvert it back. It should scramble the bytes pretty good.

The first thing youtube does, it checks audio footprint, since it is less intensive cpu task. Chaning one frame will do nothing, because unless you really do 1:1 byte copy few videos are frame identical, and they still manage to detect duplicate content.

If you have Quicktime pro - add one or two frames of random dark color along with the audio of a random sine tone. Add this to frame 2 or 3 at the front, and to the last frame in the back. Fingerprint changed.

Nope, doesn't work. Bitrate only changes "a density" of information, not so much its characteristics (even though it is affected on a scale depending how far from original bitrate you are).
