When doing keyword research then building and structuring a niche site, what is the purpose of having a main keyword?

Yes and no.
A main keyword could be the best performing keyword your site ranks for and delivers you the most traffic so you consider it to be your "main" one.
Other sites may rank for many keywords and not have a standout one so there isn't one which would be considered to be main.
Main keywords could also be for micro niche sites such as "Best 4 Slice Toaster" or something like that.

If it's a niche site as opposed to a micro niche site, can you have multiple main keywords? Are main keywords another name for seed keywords? Like the one's you'd have category pages for?

A micro niche site will have one main keyword which will most likely be in the URL of the domain name.
A niche site can have multiple main keywords which would be the ones you want to predominately target to drive traffic.

Keywords are based on pages not websites. Every page has a keyword cuz u need to make sure you willl get traffic from ranking your pages.
So, if you rank fornspecific keyword, you will rank for all of its variations but u neex at least something you know it will bring traffic to target with SEO

Another thing I always wondered was, if you're building a niche site, that has multiple main keywords, which main keyword should be in the domain name? Because usually in micro niche sites you'd buy an exact match or a partial match domain wouldn't you?
Or is it not necessary?

That one depends on your niche and there are a few things you could do.
Micro niche sites you would always buy an EMD or PMD.
For broader niche sites, you could include a closely related keyword or go for a branded name.
Example:
Let's say I sell Social Media followers. My target keywords would include the word "followers" so I would want this in my domain name.
My domain name could be igfollowers.com if I sell Instagram followers.
Or bestfollowers.com if I sold followers on multiple Social Networks.
In this example, the word "followers" is very important because it will appear in a lot of the search queries that I would be trying to rank for such as Buy Instagram Followers, Buy Cheap Twitter Followers, etc.
I'm not sure what's exactly happened with the Fred update and if it has impacted this, but before this update having the word "followers" in the domain name would have given me a benefit in the SERPS in this example.
You could include a keyword like the example above which would be in a lot of the search queries or you could include a keyword with the highest volume and that would all depend on the niche you're in.

Would a site where you sold followers for multiple social networks be considered a micro-niche site or just a niche site?

Just a niche site. The niche would be Social Media followers.
Even specializing in one Social Network like Instagram only would still be a niche - Instagram. That post above was just an example in regards to keywords.
Micro niche sites tend to be geared towards Amazon or AdSense and are your typical things like bestsomethings.com, bestsomethingreviews.com, buysomethingonline.com, etc.

Broad (( computer ram broad niche
But you also got specfic niche that got sub niche like
Plug specific niche
Plug fuse is sub of the specific niche
They all mean the same but all written in diffrent terms.
It very easy when you think a out it.
Jumper main
Tshirt under my jumper sub niche
Vest under my tshirt sub of a sub niche
Jeans main
Boxer shorts sub
Go on and on , you get the drift lol

Yes main keyword is necessary it will brings most traffic on your site. Then you will choose keyword as variation of main keyword. It will improves site traffic.
