Tier 1 links are high quality backlinks that pointing directly to the page you are trying to rank.
Tier 2 links should point directly to your Tier1 pages which in turn points to your page you are trying to rank.

When talking about the tier 1 backlinks. You will be targeting the main money website. And for tier 2, your target would switch to the links you have generated in tier 1.
