That question is too general.
If you build 2 layers of spam, it still looks like spam. If you do this right, then yes it still works.

Tiered linking works, but sadly those who say it doesn't do not actually know what it really is. Its all about flowing link juices across different layers and doing that with care.

@OP: If you're going to use wiki and crap social bookmark sites as tier 1. Then yes I'd strongly agree with you, Google hates these type of tiered link building.
I hope you get my point.
Nah man I plan to use WEB 2.0s , blog posts and article directories , for tier 1 , waht about forum signatures from high niche related PR sites , what do you guys think?

Google doesn't like any kind of link building method that involves you placing the links yourself. The only link building methods they approve of are the ones that attract links, e.g. content marketing.
Tiered linking still works, though, but it's efficacy largely depends on how it is implemented.

Tire link works.If you will not provides tire link juice then it will not provide that much juice to next tire juice.
