I am a Senuke user but I see some people do have Squidoo. Can someone tell me why people create sites like Squidoo manually? Do you create a SEO optimized one with 1 or 2 backlinks to your main site?

squidoo might help you out, its trusted site
but if you got article ready you can just register and submit it to squidoo yourself.
why pay 5$ for few minutes work

Yes, Squidoo is a web 2.0 property which is good as a tier one backlink or tier 2.

Well, the $5 would be for the article, not the process its self.

Does anyone here use Squidoo with Senuke? Also besides Squidoo, anything else?

Since im a noob i havent created any manual Web2.0s yet, i dude i know uses rankbuilder for me on my sites. i need to find a software for this sort of thing...

I have built some manually. I do not think I would pay five for one. Pretty easy to make look nice, although they can be touchy about some keywords.

I usually embed my youtube vid, add some pics and just use my spun article from my AMR run. Ten to fifteen minutes top, if I have the content ready.
