Site Disappeared for Main Keyword

too early to say anything. If it's your main money site and you can't do anything without it, I would suggest you look into other sources of traffic instead of depending on Google. Here's what to do now,

1. Start guest posting on other related/high authority blogs/sites.
2. Get your social up and running.
3. Get into niche relevant directories.
4. Participate in online forums.


Also, don't forget to sort your onpage SEO out if you did a redesign and get a feel of your keyword density.
 
My site has gone for my target keyword my main one only. It was 6/5 on the rankings but now has gone completely. What should I do?

Depending on the niche, depending on the drop whether it's penalty -50/-99/-299 or if it's just some pure flux related issues you can tell...
 
First u look in webmaster tools for any crawling errors ? and also make a try to fetch as Google bot to see what Google sees on your site ? If it all looks fine. Then Don't you think it is in Google dance? well that one always happens but you keep on doing what you and eventually it comes back with more strongish than before.
