I have a local website trying to target a UK city. However my site is a .com extension. What should I do to target this?

You can point the targeted geo. to UK in google webmaster tools. Domain extension is not so critical as you see, in my country, most of the top ranking local business sites are .com too.

I did that. Is it just the waiting to game to see if I rank?

I think it's not enough just to change your website location in Google Webmaster Tools. You need to build some backlink from UK domains as well in order to rank in UK.

I would have just got a "Cityniche.co.uk" and hosted it in the UK.

Maybe someone can shed light on this but I was also told hosting the site in the UK is as important as adjusting the web master tool, any thoughts? GeoIP of the IP should match your country your targeting.

Yes def. host it in the country you want to rank, and set WMT to UK.

One of the biggest factor in rankings is the extension in domains matching the search locale, much easier to rank a .co.uk in google.co.uk than a .com

Google Webmasters and proper backlinks for UK sites should get you closer to the top. Google says the location of your server plays a very small role in ranking, but the proximity of the server to your users can increase the speed when accessing your website: "Q: Is the server location important for geotargeting? A: If you can use one of the other means to set geotargeting (ccTLD or Webmaster Tools? geotargeting tool), you don?t need worry about the server?s location. We do, however, recommend making sure that your website is hosted in a way that will give your users fast access to it (which is often done by choosing hosting near your users)."
You can also embed google maps on your site and add it to Google+ with appropriate circles.

.COM domain names can be used to build a website tailored to customers in any country or city, so proceed when you are ready :).

I have made a change by moving my site to a .co.uk domain with my key word in it. Let the changes begin!

.com domain wont make much of a difference but where you get your backlinks from does. So, pay more attention to uk based backlinks.
