I see several people suggesting this in order to make your post look less spammy (and after a while you can also remove them without leaving traces editing the main post), does it make any difference at all for Instagram algo?

I'd like to try it but I'm a bit worried I could get penalized in the hashtags searches...

yes you could do that on your first comment.. you could even delete it and repost another set of hashtag just to make it have more engagement.. normally this method is done if you have small amount of follower and wanted to grow

I always post with these steps:

    1) Description/Call to Action (something like "Tag a friend who needs more of this in their life") with no hashtags
    2) Write the first comment with thirty hashtags (the max) immediately after (I keep a list of comments saved in notes)
    3) Write a second comment relevant to the post to bury the hashtags a little bit and make it look less spammy
