Is there any difference for WP what ranks better?
If I got just one article I want to focus on and rank would it be reasonable to create it as a page and place it in the main menu?

The point is , what are your services , how many keywords you wish to rank and if you can add all keywords to main homepage or you have to split them in pages. (That depends on how many keywords you want to try rank) if there are many then split them between pages .
This is a part of onpage optimized not offpage strategy

I am targeting just 1 KW and indeed it is onpage optimisation.
This article is supposed to serve as a ranking. E.G. "The best countries to visit" however my niche is health.
The blog mainly consists of blog posts however I am wondering whether I could make it a separate page and add to the main menu and subsequently rank it.

Hope we are on the same page.

It depends on the backlinks and how you optimize the page/posts.

They make no difference, all the same. Posts tend to be better for larger blogs with hundreds of articles, makes it easier to manage.
