Do I always have to keep that VPN on while working on X or just while tweeting?

Don't use VPN, their IPs are spammed to death. Use a proxy. And if possible, just 1 proxy per each account. 5 accounts, 5 proxies. Do you want to use the first account? Turn on the first proxy. Do you want to use the second account? Turn off the first proxy and turn the second proxy on.
