Would it matter which of these I use? Why?

MainDomain.com/Local

or

Local.MainDomain.com

What is your opinion?

There have been a lot of talking about this. For me, it's almost the same thing - it's how you backlinks/targets these subdomain/directory
 
I prefer subdomains to subfolders personally.
 
It really depends on what the sub or folder is...

Never thought about but here is a link to somebody opting for subfolders excepting for GEOs:

So I guess the logic go with a subdomain if the content and links allow it to stand alone.

What bertbaby just posted...subdomains are largely viewed as independent entities...
