What do you mean by one page affiliate site?
If you are referring to one-page sales pages; then, yes. They still exist.
As a matter of fact, the best sales pages consist of a single page.

What's your traffic source? For SEO you probably won't rank a one pager unless you get tons of reviews, social signals and traffic from elsewhere. If you can draw attention to it from somewhere other than SEO, then yes a one pager will probably convert. But you won't be able to rank it with organic SEO and no social budget.

it is possible but you have to know what you are doing in terms of SEO.., like someone said earlier, some of the best and successful are one page.

1) content including highest ranking keywords
2) product
If both are well maintained then there is no issue of page limit.
