just want to ask if profile links have to be indexed to increase serp? or it's not necessary to be indexed?

no it does not, A couple of days ago I received a 3,500 Xrummer blast to my site. The site jumped to position 1 in a matter of hours.
 
No, the links do not need to be indexed or showing in any kind of report (YSE for example). As long as the search engines (Google) are aware that they exist its good enough. That being said, I prefer to get as many indexed as possible.

Try to index mass of profile links in a short time is not a very safe idea, let google index them naturally

How do you know they're aware of the links if they're not showing up and not indexed? My logical sense has to disagree with your reasoning.
 
This is a tough one, just because Google hasn't indexed a page, it doesnt mean it doesnt know about it or doesnt count the links in it. There are several reasons for Google not to index a page. It's safe to say though, that a link from an indexed page will be worth much more than a page that hasn't been indexed.
