You can insert a validation form prior to the comment publishing, verifying that the comments are submitted by real people but not a spam.

Also you can install a plugin that makes all external links nofollow, It will help you reduce the spam since most people have their campaing setup for ******** links only
that will ensure (in most cases) that they don't spam you more than once.

Don't spam your site with unnecessary plugins. Just find above comment in your wordpress settings.
