I had to code one myself with PHP and MySQL and then use the Twitter API, to get usernames, reply and tweet.

Web based, no need to have it on your computer running 24/7: http://twitterfeed.com/ Socialoomph might be useful too, with yahoo pipes you can automate almost everything based on the criteria you choose and if you search there are comprehensive tutorials on this forum.
