What I want to know is that if I have a Amazon associate link from amazon.co.uk and someone clicks on it from the USA does it take them to the .com site and do I still get a commission if they buy even if my associate link is from the .co.uk site?

No you don't. I use Easyazon to send my customers to the Amazon in their country.

Yeah that's the problem with Amazon system. You get paid only for sales register in site you registered to. If you use Wordpress there are plenty of plugin to send visitors to right site but you need to register in all of these sites.

Amazon is a pain with this, if you have worldwide traffic and you want to earn commission from different countries then you have to register with each country or you lose that sale, if you live outside of India you cannot join that but canada, usa, france etc are ok to join.
I use autoAzon and easyazon as you can add the countries you belong to and it gets redirected automatically
