It's pretty much impossible to get any 3 letter on Instagram without buying it. Trying to turbo the username probably won't work either because most of the people that own the turbos, have very expensive ones and have also paid lots of money for the name.

Does having a .com/org/net help in your rankings opposed to using a .us/gg?

I pretty sure on instagram, if you own the .com domain, and/or you have it trademarked, you should be able to get the username if they are inactive.
