If it's literally only one video and every other video is unquestionably original content and your channel looks professional and like it represents a legitimate creator - I don't think a single copyrighted video will prevent you from being monetized.

The channel does NOT have to be approved for monetization for ads to show from copyright, but I am 99% sure it needs to meet the monetization threshold for views and subscribers. You don't make any real money before those numbers anyway.
