Are you using proxies? Make sure your proxies are not blocking Tumblr.com
Here's how you test it, load the proxies into Internet Explorer, and go to Tumblr.com and create an account. See what happens.

On proxies: When checking domain availability I'd imagine just using anon proxies would work? ie: process is going directly to the domain to check, and not using Google?
And it does not matter if "http : //" is in front of domain, or it does?

As for the other issue, the http:// shouldn't matter though it'll look nicer without.

Can you try and register those subdomains using Internet Explorer and see if it works?
I use the tool checking not availible and then I use the subdomian using Firefox and it works,have you fixed the Tumblr availability checker?
