If you redirect from one domain to another, will the spiders still index the redirected site?

No the spiders would crawl the other site.

It would redirect. Thats what redirection is for.

Is there any way to have a site ranked and then have the viewer automatically redirected to the new site? I am trying to have a site ranked and then use it to redirect.

HTML redirect.

The deal is this: I want to create a website for a friend that owns a business. He already has a site, but it would be difficult to optimize his quickly. So, I want to get a URL that matches the keywords in which his business operates and then optimize a site quickly for that URL. Then once we reach SERP 1 on G, then I want people to do their search, find the optimize site, BUT see his website. How do I do this?

What if you used an iframe the width and height of the landing page? Would the spiders only see this as one HTML element and the visitors would actually see the other site?

That's called cloaking. Be careful as this could hurt your rankings! If you do plan on cloaking your gonna have to get very creative with how you do it using IP/location based redirects as well as other techniques.

Don't just go iframing your whole site and think that will work.

yeah that could work

Milian, I hope you're not trying to f*** me up my friend! I don't take kindly to that.
Thanks for the info SuperLinks!! Now then, back to the original question....How can you create one site, optimize it and then get the traffic from that site to another site?

I use a 301 site redirect but still the site I'm redirecting to won't take it's serP place ever it just redirects so If you hoping it will swap serp status it won't but this is my experience might work for others bit I dont think it will

No, I don't want to optimize the other site. I just want to optimize a site with a keyword specific URL and have the traffic from that site redirected to the other site without effecting the new sites SERP.

Wpcloaker works for me every time just got my hands on a copy. Get it for 50 dollars with discount code. This has to be the best cloaking software out there for wp.
