Is there a difference between if I index the tier2 backlinks and not index tier2?
From my experience, if i don't index tier2 backlinks, i don't see good results,
what about you?

i second you man... Indexing is important if you want to make use of the backlink, else it doesn't really mean anything since Google wouldn't know that it exists, unless it is crawled on its own

What kind of links do you do for Tier2 links?

If its PBN links, then there is possibility those links automatically gets indexed. If its web 2 or automated links then you need to use some indexing tools.
 
tier 1 to 2 indexers, tier 2, 3 and 4 to 1 indexer. In my opinion it’s useless nowadays to build tier 2, 3, 4 links without using indexers. Except for blog comments which are already indexed. Index ratio is just a few percent without indexers. It’s like wasting money.
