I believe this can be possible, even with Su Social or TwitterDub as long as you're using pic.twitter.com to post Videos in Replies.

But if you wanna use the Upload Video function for each Reply, then you might need a custom solution yes.

You can make yourself a throwaway "storage account" and upload your videos to that account, then just take the pic.twitter.com URL of each video and save it to your Reply list.
