Will add 100+ backlink (indexes) over night for tier 1 site or tier 2 site trigger search engine attention and get penalize?
 
Depends on how much it will stand out.

What is my current link velocity? - Is it normal to have 50-70 link per day for your website? If so, 100 will not stand out that much. If you have 5-7 a month, oh yeah!

Is it typical for competitors in your niche to pull numbers like this
 
in tier 1 or 2.. i think it shouldn't trigger too much..

If you add 100+ per days, Google index only few links in day and some on next day. Not all 100 in same day. So No worries. Rather than you go Dripfeed method, Which is very safer.

The next day, Google will penalize you definitely. If you add 100s of links in a single day.

Drip feed is the best choice for building backlinks. Before choosing the site for backlinks, you must check the authority of the site.

because some link building services offer indexing. The link they build for you, will get index immediately.

At times, I manage to get lots of links (less than 50, I can't imagine getting 50+ decent links on my own in 1 day) but some links get indexed within minutes, some within days, some within weeks so it looks natural.
 
Try to target lower numbers (10-50) and keep building momentum.

From what I've observed, the effects of backlinks appear 15~30 days after you create them. Same the other way around: if you lose backlinks, your ranking will drop 1 month after the fact.

How do you decide which pages to build individual links to? Also, how about determining anchor text - or just make it look natural?

Hmmmm... with you, I smell sarcasm.
I try to mainly concentrate on keywords which are on page 2.
