Won't penalize you. Especially if the url is semantically related to the title.

In fact you're free to change the title at will to optimize it. Just don't spam, stuff, etc.

Also in the URL don't stuff it. Don't make it example.com/buy-viagra-online-prescription-cialis or something like that. Remove connecting words and irrelevant shit from URLs. Make URL as short as possible.

People do a lot more blackhat stuff than what you're doing and still don't get caught. What you're doing is totally logical, just check the search results on Google.

Also keep in mind when other questions pop out to your mind, that Google is only a smart robot and won't penalize you that easy.

What I am trying to say is, don't overthink stuff, but better do it wrong first then the second time you making sure that is correct.

What do u mean, permalink structure? I'm used to set my permalink structure on the post with the keyword. I just make sure it is not too long and not too short. I also make sure that my post title and permalink both have the similar keyword, no matter how long of your post title
