Am I allowed to post links on tweets to items on the Amazon website using my affiliate link?
Or do I have to post a link to my Niche website and have them put the Amazon link from there?

You can use direct amazon aff url. Shouldn't be an issue at all.

You can and Amazon make you shortening for you. But it's better use a Landing page rather than direct linking.
