I have several keywords to compete for. I am creating article pages in subdirectories to rank. The problem is that the way this site system is setup every time I create a subdirectory it is only accessible via /v/ so instead of visiting http://mydomain.com/keyword1 I have to use http://mydomain.com/v/keyword1. Is this going to affect the ranking?

If it's just 1-3 sub-directories, then no - it shouldn't affect the pages ability to rank at all. I've worked on some financial services websites in the past which always had at least 2 subs, as your example does, and they still achieved page 1 and top 3 rankings for highly competitive keywords.

Quick question: Can you edit the name of the "v" subfolder? For instance, could you change it to your main keyword, or something similar? (Provided you have a single word/short keyword to begin with)

It's just an extra letter and slash in the URL string - It'll make ZERO difference to any link-building.
