If you backlink a image, will your SERPs increase in Google Images?

No...an original image, with exact match keyword as filename and alt tag is the best way to go.

Google doesn't care about original images. I easily rank ahead of other images although i put mine on my site later then the others.

What If I build backlinks to where the image is located? Does that make a difference?

Google will index unoriginal images..sure, and if you use the keyword like suggested above, you may even outrank the original...but if you get a good image, that's original, it's easier to rank. Look at tineye...google can tell if an image is unique.

I really don't think backlinks make a difference...as long as your content is indexed.
