Is there any plugin/filters or method to block fake email or 10 minute emails from getting opted inside an email list, especially in case of Single Optins.

Make it double-option. Also, if you have a list of temporary email services, you can create a regex pattern and prevent users from entering emails from such domains. There are plenty of examples online on many languages such as JS, PHP, etc.. it depends on your website/requirements. If you don't want to bother with code, hire a programmer who can get it done in 10 min for cheap.

Make it implicitly stated that something will be sent to their email even if it won't. IE. "Enter your email address you wish to the receive the free report at" - using this type of language will drastically reduce BS email entries since it appears to be required 100% to complete the interaction/get something.

If you're really having a problem with it I would hire someone to edit your script like Thunder said which will blacklist popular temp email services, and also verify a domain name is registered and not jfdgnfdm32543.com - Make the error message aggressively state temporary email services are not accepted and enter their proper email address.
