That depends on too many factors... one can't make a blanket statement as to just one link without taking into consideration all the other factors.

Generally speaking it has to do with a lot more than just one link.

Onsite SEO, other backlinking you are doing, competition, etc.

Your question cannot be answered honestly without knowing all the facts.

it takes more than 20 days minimum to get indexed. One Link will not show changes in your ranking. It will just appear in google when search site:yourdomainname.com . No other use with 1 backlink, even if it is PR10. Make more and more, it will raise your Ranking in Google

It also depends how relevant that PR5 link is to your website. If the PR5 website is on the same niche as your own website it will greatly benefit it. If the link is solid, then just sit back and forget about it for now. Some links takes time to get index.

Yes it is relevant, and the page with the link was indexed sat night, about 24 hours ago. The link is on the homepage of a PR5 site and when I search for that site and look at the google cache, it was it was cached late last night. Not sure if this info helps - but thats what I know as of now.

if you are mass spamming your site you will see SERP moments quite frequently.(postive or negative)
But if you are building quality links then it takes some extra time to show up effect in SERP.

It will take around 30 days in my experience to see any action from that type of backlink.

with only one pr5, you might not see any movement, but usually a few days, try getting 10 or 20 pr5, low outbound homepage, see what happens.

This is not necessarily true. If the PR5 site where the link is coming from gets crawled very often by Google, the link will get indexed in less than 1 hour. It just depends how often the search engine engage a website. The more frequent the search engine crawl a website the quicker it will index the inner and outer links.

One link won't really do much. So don't sit infront of your PC f5ing your serps.

keep working on on-site seo, and keep building slowly.

Is the backlink on the Homepage of the PR5 or a comment link?

I think that the OP mentioned that the link is coming from the homepage of PR5 website.
