... and color vs colour affect SEO?

If I have GrayColour.com domain and I have "grey color" as main keyword, will my domain be EMD for grey color + gray colour ?

You have to target them separately. Google treats them as different search terms... i.e. "gray shoes" is a totally different query than "grey shoes". What is interesting is that this isn't always the case with synonyms. You have to check each case. Example search for "la lawyer" matches results for "la attorney". For some reason google doesn't think grey and gray are the same thing.

I also have the same opinion like "Validseo". Google normally detects the Synonyms easily but for Gray and Grey, it is better to target separately.
