After building Tier1 links and sent to indexing services, and if they are successfully indexed, do i still need to build Tier2 to those indexed Tier1 URLs?

If the Tier1 already has decent authority/traffic, I usually wouldnt bother.

If Tier1 links are on submission based sites with no traffic or low authority, I would prefer building some Tier2 links to power them.

Yes, you still may need to build Tier 2 links even if your Tier 1 links are successfully indexed, because indexing only means Google knows the page exists; it does not mean the page has enough authority (link juice) to pass significant value to your site

Indexing a Tier 1 link does not automatically mean you need Tier 2. first see whether the Tier 1 links are relevant, indexed, crawlable and actually contributing . If they're already strong placements, adding another layer just for the sake of volume may add much benefit.

Yes tier 2 is necessary if you tier 1 links are not strong enough
But for the best result I suggest use both tiers. It doesn't hurt
