Yes this can be done.
Let's say you want to look for people tweeting Rebecca Black (currently a Trending Topic) in New York.
You search like this:
Near:NY Rebecca Black
The operator after "near:" must have no spaces (like Near:NY)
You can use this for basically any state or city in the U.S.A.
Not sure how it works for other countries though...

Ok ,but i'm not looking for tweets from users from NY on a certain topic, i want to find twitter users from NY, what then?
Is there a way to search for twitter users from a certain location?
For example i want to promote a local business to local people, how could i find them?

If you search Near:NY only you'd get people from New York who are tweeting right now...
If you search Near:New York, it will not show New York tweets, instead it will find some city named "New", and show you tweets from people who are saying anything containing "york".
